The Twist Hybrid Capture workflow delivers highly enriched libraries in just 24 hours.

Users begin by setting up hybridization reactions at the bench, after which Callisto runs the overnight hybridization step. The following day, capture beads and wash buffers are loaded onto Callisto, which automates target capture, heated washes, and PCR setup. The enriched libraries are then taken off Callisto for PCR amplification, and the amplified libraries can optionally be returned to Callisto for automated post-PCR cleanup.

High quality sequencing metrics

Callisto delivers high uniformity and on-target rates comparable or better than the manual workflow. The data presented here is using the Exome 2.0 panel downsampled to 50 M reads.

Uniform coverage across targets

Callisto delivers unbiased coverage across a wide range of GC contents on par with manually processed samples. The data presented here uses the Exome 2.0 panel.

Excellent target coverage for difficult to capture regions (ACTA1and RASA4B exon)

ACTA1 and RASA4B are select examples of difficult to capture genes that are systematically undercovered. Hybridization capture on Callisto yields high depth of coverage in these gene regions of interest, with minimal or no dropouts for both high GC and low GC regions.
